The hit series on BBC, which began as a pilot radio show in 2000, beat classics from the likes of Monty Python, which stars John Cleese and Morecambe and Wise in the top 50 poll, as voted by the British public.
The winning scene from Little Britain features wheelchair-bound Andy with his gullable helper Lou at the public baths, managed to beat the much-loved Python parrot sketch to number one.
Other British comedies that made it into the top 20 were; The League of gentlemen, Goodness Gracious me, The Two Ronnies, French and Saunders and Phoenix Nights.
Little Britain has won a huge amount of high-profile awards in the last year and two places in the top five of the 50 Greatest Comedy Sketches - both centring on trips to the local pool.
In the winning sketch, viewers see Andy jump off the highest diving board while his unwitting carer Lou tells a lifeguard that his friend will need help getting into the water.
The Monty Python's Flying Circus scene, which sees pet shop assistant Michael Palin attempt to convince disgruntled customer John Cleese that his pet bird is not dead but resting, came second.
Third place went to The League of Gentlemen, for the scene when the couple running the local shop savagely murder the men who come to put a road between them and civilisation.
